If you see it sticking its paws or tail through the vent, you can try what I was able to do nearly 40 years ago in a similar situation.
I had just graduated and was living in Dallas. My mom back in Waco was getting a new roof on the house. Roofers had taken off an exhaust vent cover, and a squirrel got into the ductwork. I called home one evening and could tell from her voice she was exhausted and somewhat frantic. Squirrel had spent all night and all day running up and down the ducts with its claws echoing on the metal and running toward the light in whatever room was lit up. While we were on the phone, she screamed "It's sticking its paw through the vent!" (the vent was right above the telephone, this was 1986 so still on fixed landline). I told her to reach up and jam the vent shut to see if it would trap the squirrel. She was able to do so, so I told her to go in the kitchen and get a can of Raid and spray it directly into the vent as much as possible, which she was able to do. Next morning when the roofing crew arrived, one of them came in the house, unscrewed the vent, and removed said recently deceased varmint.
